A role for angiogenesis in rheumatoid arthritis BJMBR
Stupack,D.G.; Storgard,C.M.; Cheresh,D.A..
Rheumatoid arthritis (RA) is a chronic debilitating disease characterized by distinct autoimmune, inflammatory and fibrovascular components which lead to synovial proliferation and joint destruction. However, existing treatments specifically target only autoimmune and inflammatory components despite the fact that neovascularization of the inflamed synovium is a hallmark of rheumatoid arthritis. Angiogenesis may contribute to synovial growth, leukocyte recruitment and tissue remodeling, thus potentiating disease progression. Although no therapies currently target angiogenesis, several existing therapies have anti-angiogenic activity. Recent advances in anti-angiogenic strategies in oncology, including the identification of integrin <FONT...

Articular inflammation induced by an enzymatically-inactive Lys49 phospholipase A2: activation of endogenous phospholipases contributes to the pronociceptive effect J. Venom. Anim. Toxins incl. Trop. Dis.
Dias,Renata Gonçalves; Sampaio,Sandra Coccuzzo; Sant’Anna,Morena Brazil; Cunha,Fernando Queiroz; Gutiérrez,José María; Lomonte,Bruno; Cury,Yara; Picolo,Gisele.
Abstract Background Arthritis is a set of inflammatory conditions that induce aching, stiffness, swelling, pain and may cause functional disability with severe consequences to the patient’s lives. These are multi-mediated pathologies that cannot be effectively protected and/or treated. Therefore, the aim of this study was to establish a new model of acute arthritis, using a Lys49-PLA2 (Bothrops asper myotoxin II; MT-II) to induce articular inflammation. Methods The articular inflammation was induced by MT-II (10 μg/joint) injection into the left tibio-tarsal or femoral-tibial-patellar joints. Cellular influx was evaluated counting total and differential cells that migrated to the joint. The plasma extravasation was determined using Evans blue dye. The...

Hepatitis C virus seroprevalence and genotypes in patients with diffuse connective tissue diseases and spondyloarthropathies BJMBR
Barbosa,V.S.; Silva,N.A. da; Martins,R.M.B..
Many extrahepatic manifestations, including rheumatic diseases, have been reported to be associated with hepatitis C virus (HCV) infection. In order to investigate the prevalence of HCV infection among patients with rheumatic diseases, in the present study we interviewed 367 patients and tested their blood samples for HCV antibodies (anti-HCV) by an enzyme-linked immunosorbent assay. Anti-HCV-reactive samples were retested for confirmation by a line immunoassay and also for HCV RNA detection by the polymerase chain reaction. HCV RNA-positive samples were genotyped by INNO-LIPA. An overall HCV infection prevalence of 1.9% (7/367) was found. Of the 7 HCV-infected patients, 4 had systemic lupus erythematosus and 3 rheumatoid arthritis, resulting in positivity...

Histoplasmosis diagnosed after arthroscopy of the knee: case report BJID
Falster,Lorenzo; Marin,Maurício B.; Gomes,João Luiz Ellera.
ABSTRACTFungal arthritis is a rare complication of arthroscopic surgeries, but its possibility should always be considered due its deleterious effects on any joint. Infection caused by the fungus Histoplasma capsulatum is the most common cause of respiratory tract infections by fungi, meanwhile histoplasmosis arthritis is more rare than all other fungal infections. However, their atypical forms of arthritis and the importance of early diagnosis and treatment cannot be over-emphasized. Herein we report a case of knee monoarthritis in an immunocompetent patient with histoplasmosis arthritis following an arthroscopic meniscetomy, diagnosed by synovial biopsy and culture performed during a second arthroscopic procedure. The joint was debrided in this second...

RANK, RANKL and osteoprotegerin in arthritic bone loss BJMBR
Bezerra,M.C.; Carvalho,J.F.; Prokopowitsch,A.S.; Pereira,R.M.R..
Rheumatoid arthritis is characterized by the presence of inflammatory synovitis and destruction of joint cartilage and bone. Tissue proteinases released by synovia, chondrocytes and pannus can cause cartilage destruction and cytokine-activated osteoclasts have been implicated in bone erosions. Rheumatoid arthritis synovial tissues produce a variety of cytokines and growth factors that induce monocyte differentiation to osteoclasts and their proliferation, activation and longer survival in tissues. More recently, a major role in bone erosion has been attributed to the receptor activator of nuclear factor kappa B ligand (RANKL) released by activated lymphocytes and osteoblasts. In fact, osteoclasts are markedly activated after RANKL binding to the cognate...

Sensitivity of rabbit fibrochondrocytes to mycoplasmas BJM
Nascimento,Carlos Manuel de Oliveira; Figueiredo,Cristina Adelaide; Timenetsky,Jorge.
Primary cell culture from rabbit meniscus (fibrochondrocytes-FcrC) was infected for 24 hours with different inocula (10² to 10(7) Colony Forming Units-CFU) of Mycoplasma hominis PG-21, M. pneumoniae FH and 1428 or M. arthritidis PG-6. The severity of the different obtained cytophatic effects-CPE was inoculum, Mycoplasma species and strain dependant. These bacteria were recovered from all infected FcrC and the SP4 medium for mycoplasmas also caused toxic effect on the FcrC. It was concluded that rabbit fibrochondrocytes were sensitive to mycoplasma infection, as well as to the SP4 mycoplasma medium.

The Influence of Ca2+ on Gluconeogenesis Stimulation by Glucagon in the Liver of Arthritic Rats BABT
Kelmer-Bracht,Ana M.; Fedatto-Júnior,Zélio; Ishii-Iwamoto,Emy L.; Caparroz-Assef,Silvana M.; Bracht,Adelar.
Ca2+ participates in the stimulation of hepatic gluconeogenesis by glucagon and there is evidence that Ca2+ fluxes are modified in arthritic rats. These findings raise the question whether hepatic gluconeogenesis in arthritic rats responds differently to glucagon and Ca2+. The experimental system was the isolated perfused rat liver. In the presence of Ca2+, stimulation of hepatic gluconeogenesis by glucagon in arthritic rats was equal to that in normal rats in absolute terms, but higher in relative terms (104.5 and 45.2%, respectively). In the absence of Ca2+, however, stimulation of hepatic gluconeogenesis by glucagon in arthritic rats was smaller in both absolute and relative terms (18.5 and 41.9%, respectively). It can be concluded that the...

The use of stem cells for the treatment of autoimmune diseases BJMBR
Rosa,S.B.; Voltarelli,J.C.; Chies,J.A.B.; Pranke,P..
Autoimmune diseases constitute a heterogeneous group of conditions commonly treated with anti-inflammatory, immunosuppressant and immunomodulating drugs, with satisfactory results in most cases. Nevertheless, some patients become resistant to conventional therapy. The use of high doses of drugs in such cases results in the need for bone marrow reconstitution, a situation which has stimulated research into the use of hematopoietic stem cells in autoimmune disease therapy. Stem cell transplantation in such diseases aims to destroy the self-reacting immune cells and produce a new functional immune system, as well as substitute cells for tissue damaged in the course of the disease. Significant results, such as the reestablishment of tolerance and a decrease in...

Translation into Brazilian Portuguese, cultural adaptation and evaluation of the reliability of the Disabilities of the Arm, Shoulder and Hand Questionnaire BJMBR
Orfale,A.G.; Araújo,P.M.P.; Ferraz,M.B.; Natour,J..
The objective of the present study was to translate, adapt and validate a Brazilian Portuguese version of the Disabilities of the Arm, Shoulder and Hand (DASH) Questionnaire. The study was carried out in two steps. The first was to translate the DASH into Portuguese and to perform cultural adaptation and the second involved the determination of the reliability and validity of the DASH for the Brazilian population. For this purpose, 65 rheumatoid arthritis patients of either sex (according to the classification criteria of the American College of Rheumatology), ranging in age from 18 to 60 years and presenting no other diseases involving the upper limbs, were interviewed. The patients were selected consecutively at the rheumatology outpatient clinic of...
